Results
Project Objectives
The scientific objectives of the project are:
(O1) developing and validating a laboratory method to quantify at single-cell level the transfection efficiency by a label-free method
(O2) obtaining a functional procedure for transfection quantification independent of the cell status (synchronized or not cells) and procedural differences
(O3) implementation of a machine learning-based algorithm for an intelligent transfection efficiency assessment
The objectives specific to the Romania-Republic of Moldova collaboration:
(O4) a laboratory know-how and knowledge transfer between the researchers from the two teams
(O5) an intensification of collaboration at individual and institutional level with a better integration of Moldavian researcher into the Romanian scientific community
(O6) an increase of publications number co-authored by Romanian and Moldavian team members
Deliverables
Project Phase I
– Fully functional PC-EGT prototype (assembled, electrically tested, integrated for biological environment)
– Numerical simulations to optimize the cell culture configuration exposed to electroporation
– DHM system assembled and capable of acquiring interference images, ready for polarization module integration
– Developed internal protocols: for DHM calibration, optical component traceability and hologram acquisition
– Optimization of procedures for the preparation and study of biological samples, including the choice of parameters for future live cell experiments.
Research Stages
Project Phase I
Senior researcher Elena Achimova, Vladimir Abașkin and Veronica Cazac form State University of Moldova visited Carol Davila University of Medicine and Pharmacy during the period November 26th – December 2nd, 2025.
Outcomes of Research Stages of Phase I:
– Selection of categories of results to be prepared for scientific communications
– DHM optical configuration design: choice of off-axis architecture and detailed specifications for assembly
– DHM assembly and testing: getting a functional prototype
– Consolidated versions of documents necessary for the technical results dissemination (optical diagram, complete list of optical and opto-mechanical components, calibration protocol, etc.)
– General program of research stages foreseen for Phase II of the project
– Structure of common scientific tasks and prioritization of experimental categories
– Planning of participation in national and international scientific meetings
